Daughter of Charles d'Orléans count of Angoulême and Louise of Savoy, Marguerite was born at Angoulême April 11, 1492. Widowed in 1496, Louise lived at Amboise from 1499, choosing for her children tutors open to new ideas such as François de Moulins and Robert Hureau. At her command, Jean Thénaud wrote for them the didactic Triomphe des Vertus. Marguerite was married to Charles IV, duke d'Alençon on December 2, 1509, a union that produced no children. After the accession to the throne of her brother François I in 1515, she frequently carried out the functions of queen Claude of France. Like her mother, she was a member of the Grand Council. 

As early as 1517 she became interested in the ideas of Erasmus and the Reformation and had translations made of texts by Luther and other Reformation theologians. In 1521 her spiritual director Lefèvre d'Étaples introduced her to Guillaume Briçonnet; for three years, through an exchange of letters, Marguerite was guided along a spiritual path that she never abandoned. Under her guidance, the 'circle of Meaux', which supported the reform of the clergy and the study of the Holy Scriptures, published French versions of the first evangelical texts written in France, Germany, Switzerland and the Low Countries. She surrounded herself with spiritual advisors and preachers whom the Faculty of Theology of Paris considered heretics and blasphemers. Yet she never converted openly to the Lutheran reform.

In the King's absence, and particularly after the death of Claude in 1524, Marguerite played the role of queen and mother to the royal children. Deeply moved by the death of her niece Charlotte, she composed in her honor the Dialogue en forme de vision nocturne. Widowed following the disastrous battle of Pavia in 1525, she travelled to Spain to negotiate the freedom of her brother, taken prisoner by the emperor Charles V. She remarried on January 24, 1527; and she and Henri II d'Albret King of Navarre had a daughter, Jeanne d'Albret, and a son Jean who lived but a few months (1530). In 1529 she seconded her mother in the negotiations with Margaret of Austria that led to the Treaty of Cambrai. After the death of Louise in 1531, Marguerite played an ever more important role at court. She published the Miroir de l'âme pécheresse anonymously the same year; a second edition, that did bear her name, was briefly placed on the Index in 1533. 

When the Affair of the Placards (1534) caused the King to harden his stance against the Protestants, Marguerite and her husband returned to Béarn where they sheltered exiles. There she wrote her first Biblical and secular plays, rejoining the court only in November 1535 at Lyon. At Aigues-Mortes in 1538 she negotiated with Charles V in an attempt to recover the part of the kingdom of Navarre that he held. François I, fearing that she might be arranging the marriage of her daughter Jeanne with the heir to the Spanish throne, forbade Jeanne to leave France; and against her own will and that of her mother the princess was married to the Duke of Cleves on June 19, 1541. During these years she wrote La Coche, numerous poems, and the Triomphe de l'Agneau. At the same time she had the Decameron translated by Le Maçon and began to write the Heptameron, her sole prose work. In 1545 she obtained an annulment of her daughter's marriage and also severed relations with Calvin who had criticized her spiritual advisors. The Marguerites de la Marguerite des Princesses, a cautious compendium of her writings, was published at Lyon in 1547. While en route to Court on her brother's invitation, she learned of his death and stopped at Tusson where she wrote La Navire, a part of Les Prisons and completed the Chansons Spirituelles. 

In 1548 she took part in the marriage of her daughter and Antoine de Bourbon-Vendôme that had been arranged by the new king, Henri II. Cut off from power, the rulers of Navarre returned to their kingdom. Marguerite retreated to Odos where she died December 12, 1549, during an official trip of her husband to Paris. She was buried with the royal family of Navarre in the cathedral of Morlas. 

Published after her death and mistaken as merely a bawdy work, the Heptameron was long the source of Marguerite's celebrity. Reexamined, it has revealed political, social and religious thought similar to that expressed in the rest of the Queen's works. The scandalous anecdotes about her private life that were published up to the end of the 19th century have been discredited, as should be the image of a Marguerite lost in mystic ecstasy. Neither beast nor angel, she offered her society a frequently unflattering mirror in her didactic and propagandistic works, the importance of which has all too long been underestimated.
